This appeared to be authentic CD8, and was produced by the DC themselves, since they contained CD8 alpha mRNA. Differences between the CD11c+ and CD123+subsets in their survival in vitro have been described.24Therefore, we examined the survival of the DC subsets following 18 hours of culture. Blood BDCA-1 + CD1c + DCs were isolated using the Dendritic Cell Enrichment Set-DM (BD Biosciences), according to the manufacturer's instructions. It appears that the thymus is divided into two compartments in terms of the nature of cells expressing Ia: the cortex, in which class II antigens are exclusively expressed by endodermal epithelial cells, and the medulla, where the majority of nonlymphoid cells are Ia-positive cells of hemopoietic origin. By virtue of their activation status, DCs provide either stimulatory or inhibitory signals to lymphocytes, and thus provide a crucial nexus point by which innate and adaptive immune responses are linked.
